The sugar key

one. In the new Spider-Man movie there's a dial. Peter Parker injects RNA and powers dim. The mechanism is real: short interfering RNAs, at least seven approved medicines, silencing genes by marking their messages. The dial is fiction. The stranger part is real: every one of those medicines reaches only one room: the liver.

two. The key is a sugar: GalNAc, buckled onto the RNA like a keychain. Liver cells wear half a million locks for that sugar. The courier finds the one door in the whole house that takes its key, is pulled in. Everywhere else: no slot, no mailbox, no answer.

three. The cargo is a message whose only content: destroy the message you are reading. The courier is an instruction to erase itself and its target: a gene turned down, not off. The dial's second fiction. The body has no off switch, only a dimmer that takes days to turn, in one room.

four. Why the liver? The house's sorting office: what's foreign in the blood lands there. Here is the map of our power to change ourselves: one sugar key, one door, one dimmer that never quite goes dark. The rest of the house keeps its noise. Maybe not a limitation but a mercy: no one can switch you off from the hallway.
