Sample check: a ketamine trial write-up, line by line
This is a check, start to finish: every quoted number traced back to the primary source, verdict line by line, a link for each. Same format clients get.
The piece: Dr. Owen Scott Muir, “Something New Is Proven Useful In Bipolar Depression,” The Frontier Psychiatrists, Sep 3 2026. It covers the KET-BD trial of IV ketamine for treatment-resistant bipolar depression.
The sources: Orsini et al., JAMA Psychiatry, Sep 2 2026 (PMID 42684694); Iovieno et al. 2016 (PMID 26736040).
The check:
- Trial description: three Ontario sites, Jul 2022-Nov 2025, ages 21-65, MADRS 21+, two or more failed trials, four 40-minute infusions, 0.5-0.75 mg/kg ketamine vs 0.02-0.03 mg/kg midazolam, adjunctive meds stable throughout. Holds.
- Withdrawals: five, one ketamine and four midazolam (63/68 in the efficacy analysis). Holds.
- Allocation guess rate: “47% guessed allocation” is 31/66 in the paper. Holds.
- Response comparison: “roughly 55% vs 39%” matches Iovieno 2016 exactly (55.1% vs 39.2%, RR 1.29, 17 trials). Holds.
- Primary result: printed as “between-group difference, -7.3 points; 95% CI, -12 to 2.5.” The paper reads “95% CI, -12.0 to -2.5; P = .003; Cohen d = 0.7.” One dropped minus. As printed, the interval crosses zero, which contradicts the “significantly lower” sentence above it. That is the exact slip that becomes a screenshot saying the trial failed.
Status as of Sep 16: the line prints as written.
